Molecular Detection of Zonula Occludens Toxin (zot) Genes in Vibrio Cholerae O1 using PCR
Authors
Abstract:
Background: Zot (Zonula occludens toxin) is one of the secretion toxins of Vibrio cholerae in small intestine that binds to certain receptors in the epithelial cells and causes a change in the structure of tight junction. The purpose of this research is rapid detection of zot enterotoxin gene using PCR. Materials and Methods: The genomic DNA was extracted by DNA isolation kit and gene amplification was carried out by the zot gene-specific primers. Then, PCR products were investigated by electrophoresis on 1.2% agarose gel stained by ethidium bromide. Also, the specificity of primers was measured using bacterial samples other than V. cholerae, such as enterotoxigenic Escherichia coli )ETEC(, Salmonela. typhi and Aeromonas hydrophila. The sensitivity of the PCR reaction was also evaluated using serial dilutions of V. cholerae O1 concentration (cfu/ml). Results: The data showed that the designed primers specificity for zot gene was successful and the sensitivity of this method was determined about 142 cfu/ml. Conclusion: In conclusion, this molecular detection can be used as a simple diagnostic kit in clinical laboratories for identification of V. cholerae.

Gene encoding zonula occludens toxin (zot) does not occur independently from cholera enterotoxin genes (ctx) in Vibrio cholerae.
Of 167 Vibrio cholerae isolates screened for sequences homologous with zonula occludens toxin (zot) or cholera toxin (ctx) genes, 3.0% of non-O1, 100.0% of clinical O1, and 0.0% of environmental O1 strains contained both zot and ctx. zot was present only in strains that were ctx positive; all ctx-positive strains carried zot. The absence of zot-positive, ctx-negative strains suggests ZOT is not...

Background: Cholera is a potentially epidemic and life-threatening secretory diarrhoea characterized by voluminous watery stools, often accompanied by vomiting, and resulting in hypovolemic shock and acidosis. It is caused by certain strains of the species Vibrio cholerae which can also cause mild or in apparent infections. The aim of this study is the evaluation of Capsaicin, as a potential in...

full textDetection of RTX toxin gene in Vibrio cholerae by PCR.
A PCR that amplifies a recently discovered Vibrio cholerae RTX (repeat in toxin) toxin gene was developed. Among 166 clinical and environmental isolates of V. cholerae causing epidemics and sporadic cases of cholera in various parts of the world, all were found to be toxigenic by both PCR and HEp-2 cell cytotoxicity assay. Standard strains of the classical biotype containing a deletion within t...
full textSimultaneous Detection of Integrase and Antibiotic Resistance Genes within SXT Constin in Vibrio cholerae O1 El Tor Strains Isolated from Iran Using Multiplex-PCR
Objective(s) Amongst the various antibiotic resistant elements in Vibrio. cholerae, SXT constin (SXT-C) is important. We were going to design a quick method for determination of antibiotic resistance gene pattern in SXT-C. Materials and Methods Ninety four V. cholerae O1El Tor isolates were used in this study. Antibiotic susceptibility testing, multiplex PCR amplification of SXT-C containing...
